Two receptors, two mechanisms

The two peptides engage different levers of growth-hormone secretion in the model system.

• CJC-1295 (no DAC) — a 29-amino-acid GHRH(1-29) analogue (also called Mod GRF 1-29) with four amino-acid substitutions that improve stability against DPP-IV cleavage. Acts at the GHRH receptor.
• Ipamorelin — a synthetic pentapeptide (Aib-His-D-2-Nal-D-Phe-Lys-NH2) that acts at the ghrelin receptor GHS-R1a with high selectivity and, in reported assays, negligible effect on ACTH, cortisol or prolactin release.

Why the pair is studied together

GHRH-receptor and ghrelin-receptor activation are additive in pituitary somatotroph models: one raises the amplitude of growth-hormone release, the other triggers the pulse. Combining a GHRH analogue with a selective secretagogue is therefore a standard experimental design for studying synergistic, pulsatile secretion.
