What is NAD+?
Nicotinamide adenine dinucleotide (NAD+) is a dinucleotide coenzyme found in every living cell, composed of two nucleotides joined through their phosphate groups — one bearing an adenine base, the other nicotinamide.
What is it studied for in the laboratory?
NAD+ is used in the laboratory as a substrate and cofactor in studies of redox metabolism, sirtuin (SIRT1–7) and PARP enzyme activity, mitochondrial respiration and the NAD+/NADH ratio in cell culture and preclinical ageing models.
Mechanism
NAD+ cycles between its oxidised (NAD+) and reduced (NADH) forms as the principal electron carrier of glycolysis, the TCA cycle and oxidative phosphorylation. It is also consumed as a substrate by sirtuins, PARPs and CD38, linking cellular energy state to chromatin regulation and DNA-repair signalling.
