Research background

NAD+ cycles between its oxidised (NAD+) and reduced (NADH) forms as the principal electron carrier of glycolysis, the tricarboxylic acid cycle and oxidative phosphorylation. It is also consumed as a substrate by three enzyme families — sirtuins (SIRT1–7), poly(ADP-ribose) polymerases (PARPs) and the CD38/CD157 glycohydrolases — which links cellular energy state to chromatin regulation, DNA-repair signalling and calcium signalling [1].

In the laboratory NAD+ is used as a substrate and cofactor in enzyme assays, in measurements of the NAD+/NADH ratio, in studies of mitochondrial respiration and in preclinical models of cellular ageing where NAD+ availability is the variable under study [1][2].
