MOTS-c (mitochondrial open reading frame of the 12S rRNA type-c) is a 16-amino-acid peptide (MRWQEMGYIFYPRKLR) encoded by a short open reading frame within the mitochondrial 12S rRNA gene — one of the mitochondrial-derived peptides characterised over the last decade.

Research background

MOTS-c was described by Lee and colleagues (2015) as a mitochondrially encoded signalling peptide that inhibits the folate cycle and de-novo purine synthesis, raising AICAR and activating AMPK in cell culture and preclinical models [1]. Kim and colleagues (2018) subsequently showed that under metabolic stress MOTS-c translocates to the nucleus and regulates adaptive gene expression through antioxidant-response elements [2].

In the laboratory MOTS-c is used to study mitochondrial-to-nuclear signalling, AMPK activation, folate–methionine cycle regulation and cellular stress responses.
